A NEW NATURAL ANTIBIOTIC: TEIXOBACTIN

Antibiotic resistance is spreading faster than the introduction of new antimicrobials into clinical practice, causing a public health crisis. Most antibiotics were produced by screening soil microorganisms. This limited resource of cultivable bacteria was overmined by the 1960s. It is reported that uncultured bacteria by in vitro conditions make up approximately 99% of all species in external environments, and are a source of new antibiotics which could not be explored yet. The new technology for growing uncultured organisms by cultivation in situ or by using specific growth factors have been developed. As a result, it is reported that a new antibiotic termed as teixobactin has been discovered in the screening of the new soil bacterium named as Eleftheria terrae. Teixobactin has bactericidal effects in Grampositive bacteria inhibiting cell wall synthesis by binding to lipid II which is a a highly conserved precursor of peptidoglycan and lipid III which is a precursor of cell wall teichoic acid. It has been tested in two animal infection models: blood infections of methicillin-resistant Staphylococcus aureus and lung infections of Streptococcus pneumoniae. As a result, all animals were treated successfully and none developed resistance. If teixobactin is approved, it will be the first of a new class of antibiotics.

YENİ BİR DOĞAL ANTİBİYOTİK: “TEİKSOBAKTİN”

Ciddi sağlık problemlerine sebep olan antibiyotik direnci, etkili yeni antimikrobial maddelerin klinik kullanıma girmesinden çok daha hızlı bir şekilde ortaya çıkmakta ve yayılmaktadır. Antibiyotiklerin çoğu toprak mikroorganizmalarından elde edilmişlerdir. Bu, kültürü yapılabilen sınırlı sayıdaki toprak bakterilerileri, 1960’lı yıllara kadar yoğun olarak antibiyotik üretiminde kullanılmışlardır. Doğal ortamlarda yaşayan mikroorganizmaların yaklaşık olarak % 99’unun in vitro şartlarda kültürü yapılamamış bakteriler olduğu ve bunların henüz keşfedilememiş yeni antibiyotiklerin kaynağı olduğu bildirilmiştir.

Kültürü yapılamamış mikorganizmaları üretmek için doğal ortamlarının veya özel üreme faktörlerinin kullanıldığı yeni bir teknoloji geliştirilmiştir. Bu teknoloji ile henüz kültürü yapılamamış Eleftheria terrae olarak isimlendirilen yeni bir toprak bakterisinden, “teiksobaktin” olarak isimlendirilen yeni bir antibiyotiğin elde edildiği bildirilmiştir. “Teiksobaktin”, Gram-pozitif bakterilerde peptidoglikanın öncü maddesi olan ve çok iyi korunmuş lipid II’ye ve hücre duvarındaki teikoik asidin öncü maddesi olan lipid III’e bağlanır. Böylece, hücre duvarı sentezini engelleyerek bakterisit etki gösterir. “Teiksobaktin”, metisiline dirençli Staphylococcus aureus ile kanda ve Streptococcus pneumoniae ile akciğerlerde enfeksiyon oluşturulmuş iki hayvan modeli üzerinde denenmiştir. Sonuçta, tüm denenen hayvanlar başarılı bir şekilde tedavi edilmişler ve hiçbirinde direnç gelişimi gözlenmemiştir. Eğer onaylanırsa, “teiksobaktin” yeni bir antibiyotik sınıfının ilk üyesi olacaktır.
