Creating Transcriptome Profiling in Patients with Thromboangitis Obliterans Patients with Artificial Intelligence Modeling
Gözde Öztan, Nilgün Bozbuğa, Halim İşseverThromboangitis Obliterans (TAO), also known as Buerger’s Disease; It is a nonatherosclerotic disease peripheral vascular disease with segmental, inflammatory involvement of medium and small sized arteries, veins and accompanying nerves, the formation mechanism of which has not been clearly revealed yet. In the pathophysiology of the disease, there are findings that start with immune processes that are suggested to be triggered by tobacco exposure, and are characterized by acute inflammation and thrombus formation in arteries and veins. Since there are no clinical, radiological, laboratory and histopathological findings defined specifically for the disease, the diagnosis is determined as the clinical probability formed by combining all the data. Preferring classification methods based on artificial intelligence techniques in addition to traditional classification algorithms is valuable in terms of accurate evaluation of microarray gene expression data and guiding the diagnosis and treatment of diseases. With in the scope of this methodology study, it is aimed to develop useful biomarkers in the diagnosis, prognosis and treatment of Buerger’s Disease by applying artificial intelligence models from the transcript data obtained by the “Comparative Analysis of Transcriptome Profiles” method.With the “Affymetrix GeneChip Gene Array”, which allows the detection of transcript variants and alternative splicing events, a molecular basis will be provided for the determination of the mechanisms of Buerger’s disease of the transcripts to be investigated by evaluating the gene expression of the whole transcriptome at the gene and exon levels. At the same time, the biological processes and molecular pathways related to gene groups whose expression changes significantly in the presence of drugs will be determined by gene set enrichment analyses, and these analyzes will be performed via the “ConsensusPath” bioinformatics tool.In addition to statistical and bioinformatic analysis methods, the preference of artificial intelligence-based methods in the classification of Thrombitis Obliterans microarray data is important in ensuring linear correlations between the data.In the data set classification, it is aimed to use the artificial intelligence model based on genetic algorithm to diagnose the disease and to clarify the treatment protocol.
Yapay Zekâ Modellemesi ile Tromboanjitis Obliterans Hastalarında Transkriptom Profili Oluşturma
Gözde Öztan, Nilgün Bozbuğa, Halim İşseverBuerger Hastalığı olarak da bilinen Tromboanjitis Obliterans (TAO); orta ve küçük boyuttaki arter, ven ve yandaş sinirlerin segmenter, inflamatuar tutulumu ile seyreden, oluşum mekanizması halen net şekilde ortaya konulamamış olan, nonaterosklerotik periferik damar hastalığıdır. Hastalığın patofizyolojisinde tütün maruziyeti ile tetiklenebileceği öne sürülen immün süreçlerle başlayan; arter ve venlerde akut inflamasyon ve trombüs formasyonu ile karakterize bulgular yer alır. Hastalığa spesifik olarak tanımlanmış klinik, radyolojik, laboratuar ve histopatolojik bulgular olmadığından, tanı tüm verilerin bir araya getirilmesiyle oluşturulan klinik olasılık olarak belirlenir. Geleneksel sınıflandırma algoritmalarına ek olarak yapay zekâ tekniklerine dayalı sınıflandırma yöntemlerinin tercih edilmesi mikrodizi gen ekspresyon verilerinin doğru değerlendirilmesi ve hastalıkların tanı ve tedavisinde yol gösterici olması açısından değerlidir. Bu metodoloji çalışması kapsamında, “Transkriptom Profillerinin Karşılaştırmalı Analizi” yöntemi ile elde edilen transkript verilerinden, yapay zekâ modellemeleri uygulanarak Buerger Hastalığının teşhisi, prognozu ve tedavisinde faydalı biyobelirteçlerin geliştirilmesi hedeflenmektedir. Transkript varyantlarının ve alternatif splicing olaylarının tespitine izin veren “Affymetrix GeneChip Gen Array” ile gen ve ekzon seviyelerinde tüm transkriptom gen ekspresyonunun değerlendirilmesiyle araştırılacak transkriptlerin Buerger hastalığının mekanizmalarının belirlenmesine yönelik moleküler bir temel sağlanabilecektir. Aynı zamanda, ilaç varlığında ekspresyonu anlamlı değişim gösteren gen gruplarının ilişkili oldukları biyolojik süreç ve moleküler yolaklar gen seti zenginleştirme analizleri ile tespit edilerek, bu analizler “ConsensusPath” biyoinformatik aracı üzerinden gerçekleştirilecektir. İstatistik ve biyoinformatik analiz yöntemlerine ek olarak, Tromboanjitis Obliterans mikrodizi verisinin sınıflandırılmasında yapay zekâ tabanlı yöntemlerin tercih edilmesi veriler arasında doğrusal bağıntıların sağlanmasında önem taşımaktadır. Veri seti sınıflandırmasında genetik algoritmaya dayalı yapay zekâ modelinin kullanılarak hastalığın kesin teşhisi ve tedavi protokolünün aydınlatılması hedeflenmektedir.
